Allscripts? I'm not sure. This is very complex software that has no forgiveness for failure in a very regulated environment. Brent Shafer basically destroyed Cerner through contempt for human capital and this was compounded by the people from Oracle. I don't think old Cerner is recoverable.

I'm reliant on 1.2/1.3 300 mesh; any deviations I can't really work around. I know SubAngstrom was making 1.8/2 300 mesh some time back, I wonder if they're making other sizes now. Also, Single Particle LLC offered good pricing for ANTcryos last I asked, maybe it would be worth getting some...

Same experience! I did get some trial packs from Mitegen but they were fragile and required optimization beyond my patience. Lots of torn squares but I did get some nice particles.

I had something growing in a SDS stock solution years back and I just put it in the back of a drawer. If you want to grow in a 10% surfactant solution you deserve respect.

He's too young to understand you could be a C elegans geneticist or an architect and not someone that spends weeks freezing aggregates on rare metal disks.

I should have been clearer in my reply, I was referring to the paper in my post. I'm not a prolific author at all, but I have had demands where it was clear that nothing was enough for the reviewers. It appears that Mignot and Cambillau didn't have such demands from Nature for more experiments.

AF has been of use at a gross level for reconstructions, but for my projects the predictions have been rather poor. I don't know enough about the subject matter in the paper, but if I were to publish something similar I would only feel comfortable with complimentary experiments to prove my points.

Combining cryoEM and functional studies we found that mammalian TRPM5 exhibit unique properties and are uniquely sensitive to calcium. Coupling of MHR domains seems to influence gating, but we'll need more TRPM structures to determine this.

Sometimes I have real apprehension about the future of experimental structural biology. The predictions for my proteins of interest are currently poor and some domains are nonsensically arranged. Who is to say this couldn't change in ten, maybe five years though?
