I think there's a LOT more that can be done through transposon mutagenesis to query a genome through editing your transposon!

Like all high throughout screens we got some unusual results too, the biology of which is still to be explored ('nested' genes??)

Some of these small genes are conserved and/or neighbouring well chracterised genes. Highlights that small genes/proteins are still overlooked.

Clearly more unannotated coding potential within E. coli. Interesting to see what different methods do and don't identify.

We did this in E. coli and identified some new small proteins, validated by western blot.

PIRT-Seq: a high-resolution whole-genome assay to identify protein-coding genes Abstract. The advent of high-density mutagenesis and data-mining studies suggest the existence of further coding potential within bacterial genomes. Small

Out yesterday. Using a translation reporter on a mini-tn5 transposon, and high freq. mutagenesis, we looked at translation on whole genome scale independently of genome annotation. academic.oup.com/nar/article/...

Genetics, ecology and evolution of phage satellites Nature Reviews Microbiology, Published online: 27 March 2025; doi:10.1038/s41579-025-01156-zIn this Review, PenadΓ©s et al. explore the genetics, potential origins and life cycle of phage satellites, and they discuss the impact of these elements on the evolution of other mobile genetic elements and their host bacteria.
