Single-Molecule Studies with AFM and High-Speed AFM: From Nucleosomes to Chromosomes Atomic force microscopy (AFM) is a single-molecule imaging technique invented in 1985. Development of high-speed AFM (HS-AFM) in 2001 now allows for the direct visualization of the dynamics and kineti...

Interested in studying #nucleosomes, #chromatin, or #chromosomes at the single molecule level, atomic force microscopy (#AFM) is easy to use technique. Here is a great methods paper to guide you through the whole process. Big thanks to Keir Neuman and Yamini Dalal.

Our latest work. We identified two structurally distinct CENP-A nucleosomes at the centromere. One is associated with the CCAN and both are important for regulating centromeric transcription.

Centromere inactivation during aging can be rescued in human cells Sikder et al. uncover how centromere function is depleted during human aging. They report reduced CENP-A/C levels and centromeric transcription in aged cells. Using knockdown approaches, they identify...

The latest work from the Dalal lab. @sweta_sikder and co show how p53, CENP-B, and LSD1 can help rescue CENP-A and CENP-C levels in aged cels.
