Commentary: Christian Jorgensen @jorlab123.bsky.social discusses new study from Elhanafy et al. (https://buff.ly/4haX9PQ) which shows how identical mutations in the voltage sensing domain of sodium channels can yield differential functional effects. https://buff.ly/4jpCRUD

5/5 When stepwise reducing Nav1.9, we see the shoulder disappear. It fits to the observation that CMi-fibers have broad APs doi.org/10.1101/2024... and high expression of Nav1.9. So, the shoulder of nociceptor APs may well be supported not only by Nav1.8 but also - suprisingly strongly - by Nav1.9.

3/5 What is the role of all those Navs during AP generation? Thomas Stiehl created a computer model in which we fed HH models of Navs, generated on our patch data, according to their expression revealed by transcriptomic data published by @dianatavf.bsky.social @tedpricethepainguy.bsky.social

2/5 We find that โ not surprisingly โ Nav1.8 activates and inactivates at more depolarized potentials compared to all the other Navs. Nav1.9 โ recorded by Enrico Leipold โ and Nav1.5 activate at the most hyperpolarized potentials. Nav1.9 has hugh window currents, and prominent persistent currents.
