5/5 When stepwise reducing Nav1.9, we see the shoulder disappear. It fits to the observation that CMi-fibers have broad APs doi.org/10.1101/2024... and high expression of Nav1.9. So, the shoulder of nociceptor APs may well be supported not only by Nav1.8 but also - suprisingly strongly - by Nav1.9.

4/5 We model a CMi-fiber (sleeping nociceptor) and an Adelta-fiber. In both, Nav1.9 significantly contributed to the fast upstroke of the AP and also to the shoulder formation. We can also nicely mimic Nav related disease pathology, like erythromelalgia, in the model.

2/5 We find that โ€“ not surprisingly โ€“ Nav1.8 activates and inactivates at more depolarized potentials compared to all the other Navs. Nav1.9 โ€“ recorded by Enrico Leipold โ€“ and Nav1.5 activate at the most hyperpolarized potentials. Nav1.9 has hugh window currents, and prominent persistent currents.
