I remember when I uploaded some old pacbio data about 5 years ago it got flagged for containing 'pacbio control DNA', and was removed automatically. Which is why I was surprised so many assemblies if the ONT one made it through and got named as plasmids.

This stuff is polluting the databases because many technicians just add it because it's in the library prep protocol. That's why I bin it as soon as I receive a new kit. Are NCBI and ENA screening for this yet? If not, they certainly should! Also, do both institutions not have accounts here?!?

I just recalled some older Nanopore data and started fixing the assembly when I came across a 'weird' contig. Blasting it generated a lot of almost perfect hits to 'plasmids' all across the tree of life. Turns out it was the spike in control DNA that comes with the ONT library prep kit...

Found another instance of my plasmid/virus, this time in sequencing data of Leptographium pseudotsugae, a fungal pathogen isolated from Oregon pine (Douglas fir). We're up to 11 fungal species carrying it. Still no clue what it is πŸ€·β€β™‚οΈ
