This is a really cool transformation, congrats to all involved. Can say from firsthand experience that a lot of those existing 2-H indazole synthesis don’t work as well as advertised (at least in my hands…)

Lots more on this project that I cant share quite yet, but special thanks to the Sieber lab at TUM for hosting me in Munich to learn and perform these chemoproteomic experiments! Lipoprotein trafficking is clearly an attractive target for gram(-)’s, especially for beta-lactamase expressing bacteria.

We then demonstrated that OXA-23, the beta-lactamase that confers resistance, is itself an outer-membrane lipoprotein. Inhibiting lipoprotein trafficking via LolF thus also causes over-accumulation of OXA-23 to the inner membrane, which exaggerates these inhibitor’s antibiotic properties.

To understand this unique effect, we used a photoaffinity probe of this HTS hit to assign the Lol complex as the protein target of this compound! Resistance selection, Lol overexpression, and CRISPR studies also confirm lipoprotein trafficking as the major MOA.

Also, an awesome collaborative paper I worked on during my PhD is now published in PNAS!

Our collaborators discovered a small molecule that is a BETTER antibiotic against beta-lactam resistant bacteria than beta-lactam sensitive ones. We wanted to understand why.

IMO a poster session is way more valuable for students, esp if you’re still in the problem-solving stage of a project. I have recieved way more suggestions and useful ideas at posters than the few times I’ve given a talk.
