Check out our preprint! With new molecular mechanisms, 140 subtomogram averages, and ~600 annotated cells under different conditions, we @embl.org were able to describe bacterial populations with in-cell #cryoET. And there’s a surprise at the end πŸ•΅οΈ

Geometry was key - FIBs are flexible!

Having the ability to image the dendritic network from alternate perspectives using planar lift out gives us different insights.

In this case, molecular variabilities in the microtubule network could be teased out.

Obviously, if you want native proteins the sample needs to be vitreous.

It was a heroic effort to establish proper vitreous cryo-fixation, testing many cryoprotectants - lifting out and imaging non vitreous samples is not fun!
