ISM-FLUX offers more than just a simple experimental setup! Discover how the doughnut-shaped illumination provides a homogeneous localization uncertainty over a large FOV and learn about the self-calibrating procedure of ISM-FLUX. Check out our paper for all the details. 5/6
We tested ISM-FLUX on DNA-origami nanorulers, and achieved a median localization uncertainty below 5 nm for single molecules. This precision effortlessly resolved 40 nm and 20 nm nanorulers. 4/6
Similar to #MINFLUX, our localization uncertainty scales with L/sqrt(N), with L the TCP diameter and N the number of photons. However, while MINFLUX requires the emitter to be within the scanned area, ISM-FLUX has a much larger localization range, making it more robust. 3/6
ISM-FLUX makes use of a #SPAD array detector. This boosts localization range and offers flexibility in the scanned target coordinate pattern (TCP). We chose a circular pattern, enabled by galvanometric scan mirrors found in most LSMs. 2/6
