A Highly Reactive Cysteine-Targeted Acrylophenone Chemical Probe That Enables Peptide/Protein Bioconjugation and Chemoproteomics Analysis To date, a variety of covalent cysteine-reactive chemical probes have been reported. The most common ones include maleimide for bioconjugation and iodoacetamide alkyne (IAA) for chemoproteomics analys...

Excited to see this study by the group of @dbkonrad.bsky.social out in JACS Au.

Ortho-dichloroacrylophenone is a cysteine-directed electrophile for peptide and protein labeling as well as chemoproteomic profiling. Glad that our group could make a small contribution.

Profiling the proteome-wide selectivity of diverse electrophiles - Nature Chemistry Covalent inhibitors are powerful entities in drug discovery. Now the amino acid selectivity and reactivity of a diverse electrophile library have been assessed proteome-wide using an unbiased workflow...

How can we study target engagement and selectivity of covalent inhibitors? Which electrophilic probes are best suited to study a certain amino acid?

Our study on "Profiling the proteome-wide selectivity of diverse electrophiles" is published in Nature Chemistry.(1/7)

But that is not all! To showcase the potential of dfdc azobenzenes for the development of chemical tools, we synthesized dfdc-OptoBI-1 and demonstrated its biological activity as a red light-operated activator of TRPC6 channels in HEK293 cells.

The data was used to establish a computational approach that allows to compute realistic UV-vis spectra by combining TD-DFT excited-state calculations with MD simulations. As such, our combined experimental and computational study enables the in silico design of new highly red-shifted photoswitches.

In total, we have added 15 new visible light-operated photoswitches to the toolbox for the development of optical devices with relaxation rates across multiple orders of magnitudes and identified several examples with stronger bathochromic shifts than the dfdc azobenzene lead structure.
