Expansion-assisted imaging (3X expansion, 0.3 NA) enables brain-wide imaging with high-contrast and <400 nm effective resolution.
We were also concerned by that, so I thought we could use a cleared tissue lens and use KramersβKronig clearing to tune n to be just right.
Yeah, that doesn't seem to work... The gel shrinks quite a lot. I have a few more tricks to try before we give up, but you might also want to play with it.

Both work great on our inverted microscopes when using an FEP coverslip (www.biorxiv.org/content/10.1...).

My first, first-author paper is now out in @natphoton.nature.com! Our paper describes an iterative spectral unmixing algorithm and eight-channel camera-based hardware we developed enabling unmixing of low SNR live-cell data at video rates. www.nature.com/articles/s41...
The final version of our multispectral microscopy manuscript is now online at Nature Photonics: www.nature.com/articles/s41...
The main additions are some further comparisons to yet more unmixing algorithms, in which we find that our algorithm is still the most accurate.
