Complex genetic variation in nearly complete human genomes - Nature Using sequencing and haplotype-resolved assembly of 65 diverse human genomes, complex regions including the major histocompatibility complex and centromeres are analysed.

The second paper, also jointly led by the @embl.org and @hhu.de, used a much smaller sample set of only 65 individuals but combined several powerful sequencing methods to piece together human genomes in unprecedented detail. Both papers mark a new milestone in our understanding of the human genome.

The first, co-authored by @bernardo-rodriguez.bsky.social, discovered more than 167,000 structural variants across the 1,019 individuals, doubling the known amount of structural variation in the human pangenome. Most variants were rare, which will help accelerate the diagnosis of rare diseases.

Herpes simplex virus type 1 reshapes host chromatin architecture via transcription machinery hijacking - Nature Communications Here, the authors show how HSV-1 rewires the host cell’s chromatin architecture during lytic infection by hijacking transcription machinery and forming contacts between viral and host genomes.

The cold sore virus reshapes the human genome’s architecture, rearranging its shape in three-dimensional space so that HSV-1 can access host genes most useful for its ability to reproduce. Read full paper led by @piacosma.bsky.social in Nature Communications:

Viruses are entirely dependent on their hosts to reproduce. They ransack living cells for parts and energy and hijack the host’s cellular machinery to make new copies of themselves. Herpes simplex virus-1 (HSV-1), it turns out, also redecorates the human genome.
