Isotopically labeled internal standard peptides are unreasonably effective at establishing analytical measuring ranges for peptides liberated from tryptic digests. It’s unfortunate that IS panels are so expensive, otherwise more scientists would use internal standards to ground their measurements.

Science is more than a body of knowledge. It is a way of thinking; a way of skeptically interrogating the universe with a fine understanding of human fallibility.

If we are not able to ask skeptical questions, to interrogate those who tell us that something is true, to be skeptical of those in authority, then, we are up for grabs for the next charlatan (political or religious) who comes rambling along.
Carl Sagan

That kind of skeptical, questioning, "don't accept what authority tells you" attitude of science - is also nearly identical to the attitude of mind necessary for a functioning democracy. They have very consonant values and approaches, and I don't think you can have one without the other.

On the cover: The blood-brain barrier is a defining feature of brain tumors, shaping their biology and therapeutic response. In this issue, Bejarano et al. use single-cell RNA sequencing and spatial analysis to construct a vascular atlas of primary and metastatic brain tumors, revealing distinct alterations in endothelial and mural cells. Glioblastomas undergo substantial vascular remodeling compared to low-grade gliomas, while brain metastases exhibit even greater permeability and increased immune cell interactions. This unveiling of vascular heterogeneity is depicted as a red pencil coloring in the intricate details of tumor-associated blood vessels within a map of the human brain wherein a tumor is surrounded by diverse immune cells. This has only been possible through NIH funding.

Funding biomedical science through the NIH has been one of the greatest accomplishments of the United States of America for America, and for the world.

We are ALL better for the work that passionate scientists funded by the NIH have been able to do.

NIH ban on renewing senior scientists adds to assaults on its in-house research Policy follows firings of tenure-track scientists and suspension of training programs

Of the 4000 researchers at NIH, only 20% are tenured. They’re going to fire all the rest as their contracts come up for renewal, absolutely decimating research at the NIH. This is catastrophic, not just for Americans, but for the whole world.

A POGase is a huge advance in the glycomics field, which has typically relied on chemical or incomplete enzymatic releases (no Sia).
Expect a lot of multi-omic studies enabled by this (N-glycomics, O-glycomics, and proteomics) since the peptide backbone is left intact.
