Thrilled to share our work using live imaging to understand how Epiblast (future embryo proper) and Primitive Endoderm (future extraembryonic tissues) cell fates segregate in the preimplantation mouse embryo. Gargantuan effort led by amazing @rpkimyip.bsky.social, David Denberg and Denis Faerberg!

This could constitute a novel tissue sparing mechanism that would minimise cell elimination in cases of maternal malnutrition or low protein diets. However, this protection would be at the expense of optimal tissue function, as it would allow the persistence of dysfunctional cells in the tissue.

As the key feature of loser cells is mitochondrial dysfunction, these results suggest that cell competition is eliminating these cells when amino acids are plentiful but sparing them in protein poor
environments.

The most interesting finding from the paper is that loser cell elimination can be prevented by decreasing the levels of extra-cellular L-proline.

Sensing of extracellular L-Proline availability by the integrated stress response determines the outcome of cell competition Cell competition is a quality control acting from development to the adult that eliminates cells that are less-fit than their neighbours. How winner cells induce the elimination of losers during this ...

Very happy to share the latest preprint from the lab. We find that during cell competition, winners induce an increase in proline metabolism in loser cells, and this drives their elimination through repression of the integrated stress response.

Essential reading for all is that serve as reviewers. Please help your authors as if they were in your lab. Make your reviews constructive.

Overly onerous reviews amount to blocking each others’ work. It slows progress and demoralizes junior investigators.
