Lipid packing and cholesterol content regulate membrane wetting and remodeling by biomolecular condensates
Nature Communications - Membrane lipid packing, influenced by cholesterol, lipid chain length, and saturation, regulates the affinity of biomolecular condensates, enhancing their interactions with...
7/8 ๐ฌ Key findings:
โ
Loosely packed membranes attract condensates
โ Tightly packed membranes (higher lipid saturation, cholesterol) repel them
๐ Condensates and protein adsorption actively remodel membranes, forming tubes & double-membrane sheets
Check out the full study here:

6/8 A side note: adsorption of membrane nanotubes onto condensate can also transform them into double-membrane sheets (i.e. flat vesicles).

21.03.2025 08:46 โ ๐ 0 ๐ 0 ๐ฌ 1 ๐ 0
5/8 But thatโs not all! Condensates donโt just bind-they also make membranes tubulate. We knew that already(bit.ly/4itKMj2), but now we also see that protein binding transforms tubes into double-membrane sheets at the vesicle surface.๐ This remodeling could have implications for organelle dynamics!

Under the microscope we see 3 contact angles (and they differ from vesicle to vesicle in the sample). They are apparent as the membrane cannot exhibit a kink. From them one can calculate the real (intrinsic) contact angle
4/8 When you characterize affinity/wetting, watch out ๐! Measure not just 1๏ธโฃ microscopic contact angle (note that microscopic contact angles differ from vesicle to vesicle). Measure all 3๏ธโฃ of them to give you the real (intrinsic) contact angle.
Check also previous work ๐: link.aps.org/abstract/PRL...
21.03.2025 08:46 โ ๐ 0 ๐ 0 ๐ฌ 1 ๐ 0
3/8 So, when a condensate droplet ๐ง meets a Janus vesicle with 2 phases - say liquid ordered and liquid-disordered - it avoids the ordered/tightly packed domain and spreads on the disordered, less packed one.
You can finely map lipid packing with hyperspectral imaging &phasor analysis bit.ly/4bvw5IO

2/8 We found that membrane lipid packing-not just phase-determines interaction. Condensates favor loosely packed membranes and avoid tightly packed ones.
๐ Higher lipid packing (longer/saturated chains, cholesterol) โ lower condensate affinity ๐ซ
๐ Lower lipid packing โ higher condensate affinity โ

1/8 Exciting new work with Agu @mangiarotti.bsky.social @mpici.bsky.social ! ๐ Our latest study explores how lipid packing and cholesterol regulate the interaction of biomolecular condensates with membranes. It turns out, itโs not just about membrane phase. Packing is the key! ๐งต๐

I like your cartoon ๐ One small comment - because membranes are so soft, sessile droplets most of the time deform them and they are no longer planar. Just a detail, important mainly to membrane aficionados.
