Nope.

It's definately contaminant driven. If we load a 10th of what we'd normally load it looks fine. If we dilute the sample in the vial and load more volume - up to an equivalent on-column loading of protein in some cases, that also looks fine. πŸ€¦β€β™‚οΈ

Ah now that we've not tried. Other samples, including pure versions of this peptide, our BSA QC and an old (degraded but workable) sample from this study look OK so I think LC is sound and it's something in the sample(s) but Ive no idea what....

And the second peak is not the contaminant, which.is what I thought at first - we see this on several (not all) peptides. Light and heavy.

Not that we can see. Middle of gradient. 1st peak stays static w.r.t.intensity despite increased loading, when we load double the second peak appear with similar area (but lower height/wider) and as we increase load only the second new peak gets bigger. Thinking contaminant in sample but it's... odd

Yup. If we run heavy only on same method it looks perfect. Guess is something in the sample (looks fine with a low load but gets worse as we load more fits with that too). But give no idea what. I've run a LOT of wierd clinical samples - not as wierd as some of yours! - but never seen this...

Everything looks great if you run heaviest only. It's gotta be something in the sample. Not a clue what though...
