Using Dark Dyes for Fast Super-Resolution Imaging: A Proof-of-Concept Study
DNA-PAINT (Point Accumulation for Imaging in Nanoscale Topography) exploits the transient hybridization of a short fluorescent DNA imager strand to a complementary docking strand to achieve super-reso...
Can one use 'Dark' or non-emissive dyes for #fast #Super-Resolution #fluorescence π #microscopy?
In this work, we show the first proof-of-concept of Dark-RET (DRET) #DNA-PAINT, using a dark donor with a bright acceptor, for cellular imaging in <30 seconds. Link β¬οΈ

The known unknowns of the Hsp90 chaperone
An exploration of the unanswered questions in how the molecular chaperone Hsp90 supports protein homeostasis, and how single-molecule techniques could drive future breakthroughs in answering them.
What a way to start 2025! Our review on the Known Unknowns of the Hsp90 Chaperone has been published!
Huge thanks to Laura Silbermann and Benjamin Vermeer for all of their efforts, and to @scisonja.bsky.social for the great collaboration :)

Fluorogenic binders would enable DyeCycling on regular coverslips β instead of the zero-mode waveguides that we use so far. We discuss the pros & cons of a range of fluorogenic systems in the context of #DyeCycling and #smFRET experiments.
Comments welcome!
