As Fran was nice enough to highlight this, I thought I'd put up a brief thread. 3 in 10 proteins that our cells make are either embedded in membranes (ion channels, adhesion molecules, etc.) or secreted (insulin, antibodies). They move through the secretory pathway.

Membrane editing with proximity labeling reveals regulators of lipid homeostasis - Nature Chemical Biology Coupling an optogenetic lipid-modifying enzyme with proximity labeling reveals protein networks and mechanisms regulating lipid homeostasis in the membranes of target organelles.

Thrilled to share our latest study, led by @reikatei.bsky.social, in @natchembio.nature.com! We began by asking a simple questionβ€”how do cells know if they have too much of a lipid in a particular membrane, and how do they respond to rectify this imbalance?

With @tomtom-auer.bsky.social team, we asked how #evolution reshapes what animals #eat to match their ecological niches. Using pan-neuronal Ca2+ imaging, we show that the changes are in how the brain processes #taste.

Also, I aliquot into 20-50ul and then freeze thaw. I haven't noticed issues with that (but also I don't freeze thaw more than a couple of times).

For yeast and bacteria I used it at 1:100, and that seemed to work well. I noticed that for mouse tissue it needs to be more concentrated.

It's supposed to be 200X, but for yeast and bacteria I treated it like 100X. When I use mouse tissue I use it a little more concentrated (some slight differences in concentration depending on which mouse tissue)
